TapHi-C for profiling genome-wide chromosome conformation capture

Abstract

Chromosomes are highly packed in the cell nucleus, with each chromosome occupying a specific nuclear territory. High throughput chromosome conformation capture (Hi-C) has been used to reveal hierarchical chromatin architectures (i.e., compartments, topologically associating domains, and chromatin loops), but requires large amount of tissue and deep sequencing coverage and is only able to provide a limited resolution in plants. However, accessing high-resolution interaction information is critical for better disclosure of chromosomal architecture spatial configuration changes under various stresses in plants.
