Light Scattering for Biopolymer Characterization

  Objective: To provide an introduction on how static and dynamic light scattering detectors determine absolute molar mass, size, and conformation of biopolymers in solution. Various applications and new tools including protein conjugate analysis, field flow fractionation, and automated DLS will be discussed.

  Speaker: Dr. Michelle Chen, Head of Analytical Services at Wyatt Technology, graduated from the Chemical Engineering Department, Yale University, under the advisory of late Professor Csaba Horvath. At WTC, she has been working with polymer characterization using different light scattering techniques in conjunction with various branches of HPLC and FFF.

  Agenda:

  Introduction to light-scattering theory 15 min

  How absolute molar mass and size are determined

  Flowing mode – on-line MALS and QELS

  MALS-UV-RI: measuring extinction coefficient and protein conjugates

  Batch mode - use of flow cell, scintillation vials and microCuvette

  Polymer characterization using MALS/QELS detection 10 min

  Measure absolute MW and hydrodynamic radius

  Determine oligomerization states of proteins and aggregation

  Provide preliminary structural analysis

  Characterize protein conjugates, membrane proteins

  Obtain size/distribution information of micelles and liposomes

  Example applications 20 min
