聚焦纳米酶仿生设计及新特性的探索,发现铁硫纳米酶具有多种独特的生物学活性,基于纳米酶建立仿生抗菌新策略,并推动转化应用。
1. 纳米酶的理性设计和特性拓展。首次报道了四氧化三铁过氧化物纳米酶后,仿照天然酶活性中心结构理性设计纳米酶,提高催化活性超越天然酶,发现纳米酶的多酶活性和低温催化特性,提出纳米酶过氧化物酶体,人工模拟亚细胞器复杂功能;发现Aβ蛋白聚集体是一种天然纳米酶,不仅与AD的发生相关,还将生物催化体系从蛋白酶、核酶拓展到纳米酶。
2. 铁硫生物活性材料。发现纳米铁硫材料(FeS2, Fe3S4, Fe7S8, FeS等晶体或MOF类结构)能够模拟铁硫簇功能,具有多种氧化还原酶类活性,其中亚稳态硫化铁还能够响应性释放亚铁-多硫活性物质,不仅调控氧化还原代谢、铁死亡,还能通过多硫化修饰影响蛋白功能,在抗肿瘤、抗菌、抗病毒、抗氧化方面具有潜在的应用价值。
3. 纳米酶仿生抗菌新策略。提出基于纳米酶人工模拟免疫细胞溶酶体多酶协同杀菌原理,快速、高效清除多种耐药菌、生物膜(Biofilm)和胞内菌,发现并阐明其介导的细菌铁死亡机制,并将这一杀菌机制拓展到病毒和真菌,在此基础上提出纳米酶抗生素(Nanozybiotics)新概念,在口腔龋齿、表皮伤口、肺炎、肠炎、胆囊炎、附睾炎、阴道炎、败血症等多种感染模型中验证了显著的防治效果。
4. 纳米酶转化应用。在口腔龋病、细菌性阴道炎防治方面开展临床相关研究,专利转让宝洁等公司;建立纳米酶小试-中试-放大生产工艺,生产规模达到吨级,,同时作为骨干共同制定多项纳米酶相关国家标准和国际ISO标准,为纳米酶产业转化奠定基础。
