生命离不开氧,细胞的氧化还原状态直接调控蛋白质等大分子功能,氧化还原应激是细胞信号转导和效应的重要调控方式,介导衰老、神经退行性疾病、代谢病、肿瘤等许多生理和病理过程。陈畅课题组通过研究生物大分子特异氧化还原修饰,尤其是巯基亚硝基化修饰的研究,回答了细胞氧化还原调控的特异性问题,并阐释了氧化应激的信号作用分子机制和生理功能,揭示了亚硝基化修饰调控"年老忘事"和"中年发福"的新机制;提出精准氧化还原概念和"5R"精准氧化还原调控原则(Right time/place/level/target/species),破解了半个多世纪以来抗氧化药物研发僵局,开启精准氧化还原医药研发;提出了"通过增大氧化还原信号阈值RST、提高氧化还原应激响应能力RRC、延缓氧化还原应激响应抵抗RRR"的主动抗衰老主动健康策略,提出"抗衰老要从小做起";系统解析了《本草纲目》记载的枸杞子 "久服坚筋骨,轻身不老,耐寒暑,易颜色,变白" 传统功效的科学内涵和分子机制,获得了明确靶点、明确有效成分、明确分子机制的衰老干预策略。
