  糖尿病是危害人类生命健康的一种重大代谢性疾病,我国是世界糖尿病第一大国,已成为我国公共健康面临的最大挑战之一。我们团队围绕胰岛素分泌的分子机制开展了系统的研究,近期主要以机体中分泌胰岛素的胰岛器官为研究对象,通过建立和利用胰岛类器官模型,研究人类胰腺的发育机制和糖尿病的发病机制。具体研究内容包括:

1. 胰岛素囊泡转运和分泌的分子机制

  以多个关键调控蛋白为切入点,利用生化和分子生物学、细胞生物学、蛋白质组学、基因编辑、活细胞超分辨成像、干细胞和类器官、动物模型等手段,研究胰岛素囊泡在胰岛beta细胞中生成和成熟、转运和分泌、以及内吞和再循环过程的分子机制。

2. 胰岛类器官模型的建立和应用

  利用干细胞、基因编辑、生物材料等多种技术,发展胰岛类器官的诱导方法,获得结构和功能更成熟的胰岛类器官模型;并应用该模型,研究胰岛的发育、组成和功能,以及糖尿病相关基因异常造成胰岛发育缺陷和功能障碍的分子机制及其导致糖尿病的致病机制。
