高时空分辨解析染色质动态结构变化,是定量描述在遗传信息的读取、复制、维护过程中染色质定向调整机制的关键,也是连接染色质结构和功能的关键桥梁。精确解析生物大分子之间相互作用及动力学行为是单分子操控技术的优势,为准确描述染色质高级结构及其表观遗传调控机理打开了一个新的窗口。我们课题组开发并利用磁镊、光镊、AFM及单分子荧光等多种单分子技术手段,在染色质纤维中四核小体单元动态组装机制、组蛋白分子伴侣、组蛋白泛素化修饰、染色质重构因子对染色质结构及手性调控机制的方面取得系列进展。未来我们课题组将发展多种体外单分子表征技术,构建核小体及染色质纤维等高级结构动态结构和调控的全景图像,理解染色质错误组装所产生的疾病的分子机制。同时,我们将开发体内染色质实时操控的技术手段,跟踪DNA复制、基因转录等关键生命活动的动态过程。
